Description:
The 23C6 monoclonal antibody reacts with the human CD51/CD61 dimer, also known as integrin αv/β3. CD51 (~120 kDa) can associate with other β subunits such as β1 (CD29), β5, and β6 to form receptors for extracellular matrix components. CD51/CD61 heterodimers are expressed by melanoma cells, endothelial cells, and osteoclasts, and at low levels by platelets. The complex mediates adhesion to fibrinogen, fibronectin, vitronectin, and thrombospondin.
Applications Reported:
Reported for use in flow cytometry and immunohistochemistry (frozen tissue). Also used in blocking adhesion processes. (Use Functional Grade purified 23C6 for functional assays.)
Applications Tested:
Tested by flow cytometric analysis of human melanoma cell lines and peripheral blood. Recommended ≤1 µg/test (final volume 100 µL). Cell number may vary (10⁵–10⁸ cells/test). Titrate for optimal performance.
Target Information
CD51 and CD61 (integrin αV and β3) form a complex that binds vitronectin at the RGD sequence and can also bind fibrinogen, von Willebrand factor, thrombospondin, fibronectin, osteopontin, and collagen. Reactivity is observed with osteoclasts, placenta, melanoma cell lines, and weakly with platelets.
