Description:
The 9F10 monoclonal antibody reacts with human CD49d, the 150 kDa integrin alpha 4 subunit. CD49d forms a complex with integrin beta1 (CD29), known as VLA-4, which functions as a receptor for fibronectin and VCAM-1 (CD106). This complex is expressed by thymocytes, peripheral lymphocytes, monocytes, and eosinophils. CD49d also associates with integrin beta7 and binds to Mucosal Addressin Cell-Adhesion Molecule-1 (MadCAM-1).

Applications Reported:
The 9F10 antibody has been reported for use in flow cytometric analysis, immunohistochemical staining, and in vitro functional assays. For functional assays, use the Functional Grade purified 9F10 (Product #16-0499).

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use ≤0.25 µg per test (100 µL final volume). Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information

Integrin alpha 4 (CD49d, ITGA4) belongs to the integrin alpha chain family. Integrins are heterodimeric membrane proteins composed of alpha and beta chains. The alpha4 chain associates with either beta1 or beta7. Ligands include MadCAM, VCAM, and fibronectin. MadCAM is the principal ligand for integrin α4β7. HIV-1 envelope can mimic MadCAM by binding and signaling through integrin α4β7, the gut mucosal homing receptor for peripheral T cells.
