Immunogen Phospho-Ago2 (Ser-387) synthetic peptide (coupled to KLH) corresponding to amino acid residues surrounding serine 387 in human Ago2. This peptide sequence is highly conserved in mouse, rat, chick, and fish Ago2. The site is also conserved in Ago1 and Ago4, but the homology surrounding the site is low between Ago2 and these two family members.

Product Specific Information

This antibody was cross-absorbed to unphospho-Ago2 (Ser-387), before affinity purification using phospho-Ago2 (Ser-387) peptide (without carrier).
The antibody detects a 97 kDa protein corresponding to the apparent molecular mass of phosphorylated Ago2 on SDS-PAGE immunoblots of human A431 cells treated with Calyculin A and mouse recombinant Ago2 full-length protein.
This reactivity is not observed after lambda phosphatase dephosphorylation.


Target Information

Ago proteins are broadly expressed in somatic cells, associate with miRNAs, and are key actors in RNA silencing pathways.
Only Ago2 displays endonucleolytic or “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when base-pairing is perfect.
With partial complementarity, Ago2 represses translation instead of cleavage.
Ago2 plays a non-redundant role in RNA interference, translational repression, and heterochromatinization.
As a core element of the RISC complex, AGO2 initiates degradation of target mRNAs through its catalytic activity guided by siRNAs or miRNAs.
Overexpression of Ago2 has been linked to tumor cell growth and cancer prognosis.
Gene disruption studies in mice demonstrate Ago2’s essential role in embryonic development and regulation of B-lymphoid and erythroid development.
