Product Specific Information
This antibody was cross-absorbed to unphospho-Ago2 (Tyr-393) before affinity purification using phospho-Ago2 (Tyr-393) peptide. It detects a 97 kDa protein corresponding to phosphorylated Ago2 on SDS-PAGE immunoblots of human A431 cells treated with EGF. This reactivity disappears after lambda phosphatase dephosphorylation.
Target Information
Ago proteins are broadly expressed in somatic cells and associate with miRNAs, acting as key mediators in RNA silencing pathways. Among them, Ago2 uniquely possesses endonucleolytic “Slicer” activity, enabling miRNA-directed cleavage of target mRNA when perfect base-pairing occurs. With partial complementarity, Ago2 represses translation instead of cleavage. Ago2 plays crucial roles in RNA interference, translation repression, and heterochromatinization. As part of the RISC complex, AGO2 initiates mRNA degradation guided by siRNAs or miRNAs and also regulates miRNA maturation. Overexpression of Ago2 is linked to tumor growth and cancer prognosis. Mouse gene disruption studies confirm its essential role in embryonic development and B-lymphoid and erythroid regulation.
