The eBioHVEM-122 antibody reacts with Herpes Virus Entry Mediator (HVEM, TR2), a member of the TNF-receptor superfamily. HVEM is expressed on various cell types including T cells, B cells, monocytes, neutrophils, and dendritic cells. It functions as a cellular mediator for herpes simplex virus (HSV) entry through binding of HSV envelope glycoprotein D (gD), which is part of the viral entry mechanism.

The cytoplasmic region of HVEM binds to several TRAF family members, mediating signal transduction pathways that activate immune responses. HVEM also serves as a ligand for BTLA (B and T lymphocyte attenuator), and the conserved BTLA-HVEM interaction between mouse and human indicates its importance in regulating lymphocyte activation and immune homeostasis.

Applications Reported: Flow cytometry and immunoblotting.
Applications Tested: Tested on normal human peripheral blood cells for flow cytometry at ≤0.25 µg/test (100 µL final volume). Optimal antibody titration is recommended.

Target Information

TNFRSF14 (HVEM) is a member of the TNF-receptor superfamily and acts as a mediator of HSV entry via binding to viral glycoprotein D. The cytoplasmic region interacts with TRAF family proteins to activate immune signaling pathways. In T cells, TNFRSF14 activation promotes proliferation and cytokine production, enhancing CTL-mediated tumor immunity. Multiple alternatively spliced transcript variants have been described, though some full-length forms remain undetermined.
