- Reconstitute with 1 mL of distilled water.
- After reconstitution, store product undiluted at 4ºC in the dark.
- For FACS analysis, use 10 µL of the suggested working dilution to label 1×10⁶ cells in 100 µL.
- Mouse anti-Human CD40 antibody, clone LOB7/6 recognizes the human CD40 cell surface antigen, a 48 kDa glycoprotein expressed by B lymphocytes and weakly by some monocytes.
Target Information
CD40 antigen is a single-chain glycoprotein belonging to the tumor necrosis factor/nerve growth factor superfamily. It shows significant homology to the Hodgkin’s disease-associated antigen CD30. CD40 is expressed on all B cells except plasma cells and is also found on some epithelial cells, carcinomas, and lymphoid dendritic cells. It plays an essential role in immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
AKNA transcription factor regulates CD40 expression, facilitating homotypic cell interactions. TNFR2 adaptor protein interacts with CD40 to mediate signal transduction. The CD40-CD40L interaction is necessary for amyloid-beta-induced microglial activation, implicated in early Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
