Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. CD14 serves as a receptor for LPS-LBP complexes, binding with high affinity to monocytes.

Applications Reported:

• Flow cytometric analysis
• Immunohistochemical staining of formalin-fixed paraffin-embedded human tissue
• In vitro functional studies (functional grade recommended for assays)

Applications Tested:

• Flow cytometry: ≤ 0.5 µg/test (100 µL sample volume, 10⁵–10⁸ cells/test)
• IHC (Paraffin): ≤ 20 µg/mL (low pH antigen retrieval recommended)
  Antibody should be titrated for optimal performance.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a lipopolysaccharide receptor and is released into serum as soluble CD14 (sCD14). CD14 interacts with LPS-binding protein (LBP) to mediate immune responses and plays roles in inflammation, infection, and allergy. Alternative splicing yields multiple transcript variants encoding the same isoform. Associated diseases include Mycobacterium chelonae infection and croup.
