Product Specific Information

The eBioCB16 monoclonal antibody recognizes CD16 (Fc gammaRIII), the low-affinity receptor for IgG with an apparent molecular weight of 50–80 kDa. CD16 is represented by two genes, CD16A (FcγRIIIA) expressed on macrophages and NK cells, and CD16B (FcγRIIIB) expressed on neutrophils.
Two polymorphisms of CD16B, NA-1 and NA-2, are known; individuals homozygous for NA-2 exhibit lower phagocytic capacity. CD16 preferentially binds IgG1 and IgG3 and minimally binds IgG2 and IgG4. Upon IgG binding, CD16 initiates signaling cascades leading to ADCC, phagocytosis, degranulation, and proliferation.

Applications Reported: Suitable for flow cytometric analysis. Binding of eBioCB16 is not blocked by MEM-154 human CD16 antibody.
Applications Tested: Tested on normal human peripheral blood cells at ≤0.5 µg/test (100 µL final volume). Optimal titration recommended.

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance, antibody-dependent cellular cytotoxicity, and viral infection enhancement. The gene FCGR3A shares high similarity with FCGR3B on chromosome 1.
FCGR3A is expressed on NK cells as a transmembrane glycoprotein, while FCGR3B is expressed on neutrophils via PI linkage. Mutations are associated with Immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and alloimmune neonatal neutropenia.
Alternative splicing produces multiple isoforms.
