Target Information

The Suppressor of Cytokine Signaling (SOCS) and cytokine-inducible SH2 proteins are a family of intracellular proteins that regulate immune cell responses to cytokines.
SOCS1 suppresses dendritic cell (DC) and T cell hyperactivation following cytokine signaling by inhibiting JAK tyrosine kinase, which is required for type I and II cytokine receptor signaling. SOCS1 binds directly to the catalytic domain of the kinase and also possesses E3 ubiquitin protein ligase activity, leading to polyubiquitination and degradation of target proteins by the proteasome.
Through this mechanism, SOCS1 negatively regulates signaling by Toll-like receptors TLR2 and TLR4 via degradation of the TLR signaling adaptor protein TIRAP.
