Target Information

FXR (farnesoid X receptor; NR1H4) belongs to the nuclear receptor superfamily of ligand-activated transcription factors (NR1 subfamily). It acts as a receptor for bile acids such as chenodeoxycholic acid, lithocholic acid, and deoxycholic acid.
FXR plays an essential role in bile acid homeostasis by regulating genes involved in bile acid synthesis, conjugation, and enterohepatic circulation.
Additionally, FXR regulates lipid and glucose homeostasis and participates in innate immune response. The FXR-RXR heterodimer binds to FXR response elements (FXREs) with inverted repeats of the consensus sequence 5'-AGGTCA-3' (IR-1) and can also bind to DR1 sites with lower affinity.
In the liver, FXR activates transcription of NR0B2, indirectly inhibiting CYP7A1 and CYP8B1, and regulates SLC10A1/NTCP, SLC27A5/BACS, BAAT, ABCB11/BSEP, ABCC2/MRP2, and ABCB4, which are involved in bile acid transport and metabolism.
