• Specificity: Clone 3.2.B8 recognizes an epitope on mouse/human PD-L2 and binds both species equally well.

• Product Preparation: Functional grade preclinical antibodies are manufactured in an animal-free facility using in vitro, protein-free cell culture techniques. Purified via a multi-step process including protein A/G to ensure low endotoxin, minimal leachable protein A, and low aggregate levels.

Target Information

Programmed death-ligand 2 (PD-L2, also known as B7-DC) is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1, an inhibitory immune checkpoint receptor. PD-L2 is primarily expressed on dendritic cells but can be induced on various immune and non-immune cells depending on environmental conditions.
Expression is upregulated by cytokines such as IL-4, TNF-α, and IFN-γ. PD-L2 binds PD-1 with 2–6 times higher affinity than PD-L1, contributing to immune regulation by inhibiting T-cell activation, proliferation, and cytokine secretion (e.g., IFN-γ, IL-10).
Recent findings indicate PD-L2 also interacts with repulsive guidance molecule b (RGMb), influencing respiratory tolerance via BMP and neogenin signaling. The native human PD-L2 consists of a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
