Target Information
The Pax genes are a family of transcription factors active in specific tissues during early embryonic development. Pax family members possess a DNA-binding domain encoded by the paired box, enabling them to regulate genetic programs. Several Pax genes are associated with developmental mutations, such as:
- Pax-3: associated with Waardenburg syndrome
- Pax-6: associated with Aniridia
- Pax-2: associated with Wilms tumor
During embryogenesis, Pax-2 is expressed in the developing kidney, particularly in condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Pax-2 mRNA and protein levels decrease as the tubular epithelium matures. However, Pax-2 expression persists in the undifferentiated epithelium of Wilms’ tumors, correlating with the proliferation of poorly differentiated epithelial cells. Expression of the Wilms’ tumor suppressor protein WT1 coincides with down-regulation of Pax-2, suggesting WT1 can directly repress Pax-2 transcription.
