Product Specific Information
This antibody is predicted to react with non-human primate, mouse, and rat based on sequence homology.
On a non-reducing gel, intact IgG appears as a ~150 kDa band, and upon reduction, generates a ~25 kDa light chain and a ~50 kDa heavy chain.
Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems by cloning specific antibody DNA sequences from immunoreactive rabbits. Individual clones are screened to select optimal candidates for production.
Advantages of recombinant rabbit monoclonal antibodies:
- Enhanced specificity and sensitivity
- Lot-to-lot consistency
- Animal origin-free formulation
- Broader immunoreactivity due to larger rabbit immune repertoire
Target Information
IL-21 is a 17 kDa immunomodulatory cytokine produced mainly by Natural Killer T (NKT), T helper 17 (Th17), and T follicular helper (TFH) cells.
In TFH cells, IL-21 promotes autocrine signaling via the IL-21 receptor (IL-21R) and STAT3, leading to transcriptional activation by Bcl6.
IL-21 is critical for TFH cell development, T cell-dependent B cell differentiation, IgG production, and induction of apoptotic signaling in naive B cells.
In Th17 cells, IL-21 expression and feedback through STAT3, IRF4, and RORγt upregulate IL-23R, facilitating Th17 maturation and maintenance by IL-23. IL-21 also downregulates Foxp3 expression.
IL-21 deficiency in mice confers resistance to chemically induced colitis, while elevated IL-21 levels are associated with colitis models.
