Target Information

iNOS (Inducible Nitric Oxide Synthase, NOS) is an enzyme responsible for the production of nitric oxide (NO), a gaseous free radical involved in cell signaling. NO mediates vasorelaxation, neurotransmission, and cytotoxicity. iNOS expression is inducible in the liver and other cell types such as macrophages, hepatocytes, synoviocytes, and smooth muscle cells upon stimulation by cytokines (e.g., IFN-γ, TNF, IL-1, IL-2) or lipopolysaccharides (LPS).

NOS enzymes catalyze the oxidation of L-arginine to produce L-citrulline and NO. The NOS family includes three isoforms:

  1. nNOS (type I) – neuronal
  2. eNOS (type III) – endothelial
  3. iNOS (type II) – inducible

All NOS isoforms contain calmodulin, NADPH, FAD, and FMN binding domains. Cytokine induction of iNOS results in delayed but sustained NO production. iNOS dysfunction has been linked to diseases such as achalasia and impotence.
