Product Specific Information

The eBioH4B4 monoclonal antibody reacts with human CD107b (LAMP-2), a highly glycosylated type I transmembrane protein (~105 kDa) localized mainly in lysosomal and endosomal membranes. It is also found on the surface of degranulating T cells, activated platelets, and certain cancer cells. Mutations in CD107b are associated with Danon disease, a lysosomal glycogen storage disorder.

Applications Reported:
Purified anti-human CD107b (LAMP-2) is suitable for flow cytometric analysis and surface staining in degranulation assays. Fluorochrome-conjugated formats are recommended for flow cytometry.

Applications Tested:

• Intracellular staining and flow cytometric analysis of Jurkat cells (≤0.5 µg/test).
• Immunoblotting at 1 µg/mL.
  Antibody concentration should be optimized for each assay.

Target Information

LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a key lysosomal membrane protein involved in lysosome biogenesis, fusion with phagosomes, and regulation of autophagy (particularly chaperone-mediated autophagy).
It is a glycoprotein (~100–130 kDa) with a large luminal domain, a transmembrane region, and a short cytoplasmic tail.
LAMP2 has three isoforms (LAMP-2A, -2B, -2C) and, together with LAMP1 (CD107a), is essential for proper lysosomal function.
Deficiency in LAMP2 leads to autophagosome accumulation and is linked to Danon’s disease.
