Product Specific Information

The 122 monoclonal antibody reacts with mouse B7-DC, also known as PD-L2. B7-DC is a member of the B7 family with a predicted molecular weight of ~25 kDa and belongs to the immunoglobulin superfamily. It has a short cytoplasmic tail (4 amino acids) and is primarily expressed by dendritic cells and monocytes/macrophages in mice.

B7-DC does not bind CD28/CTLA-4 but serves as a ligand for PD-1. The PD-1 and B7-DC/PD-L2 interaction can either co-stimulate or suppress T cell proliferation depending on the activation state. The 122 clone blocks binding of TY25 (Product #14-5986).

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on bone marrow-derived dendritic cells at ≤1.0 µg/test (in 100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Fluorophore details:
Super Bright 436 is excited by the violet laser (405 nm) and emits at 436 nm. Recommended detection filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401) to minimize non-specific polymer interactions.

Target Information

Programmed death-ligand 2 (PD-L2, B7-DC) is a member of the B7 ligand family and a ligand for PD-1. It is mainly expressed in dendritic cells but can be induced in various immune and non-immune cells. PD-L2 expression increases under IL-4, TNF-α, and IFN-γ stimulation.

PD-L2 binds PD-1 with higher affinity than PD-L1 and plays a role in inhibitory immune checkpoint signaling, reducing T cell activation and cytokine secretion. It can also stimulate T cell proliferation via other receptors, such as RGMb, which participates in respiratory tolerance regulation via BMP/neogenin pathways.
