Description:
The monoclonal antibody OX108 recognizes human CD200R (also known as OX2). CD200R is an inhibitory receptor structurally similar to its ligand CD200 but lacks a typical ITIM domain. Instead, its cytoplasmic domain contains a phosphotyrosine binding domain (NPXY) that interacts with SHIP to inhibit ERK, JNK, and MAPK p38 pathways, leading to suppression of TNFα secretion in activated macrophages. CD200R isoforms are implicated in immune tolerance, particularly in tolerogenic dendritic cells.

Expression is restricted to hematopoietic cells, including myeloid cells (monocytes, macrophages, dendritic cells, neutrophils, mast cells, and basophils), subsets of T lymphocytes, Langerhans cells, and dendritic epidermal T cells. The OX108 epitope is thought to be near the CD200 binding site.

Target Information

CD200R (OX2R) is a receptor for the OX-2 membrane glycoprotein. Both receptor and ligand are cell surface glycoproteins with two immunoglobulin-like domains. CD200R is mainly expressed on myeloid lineage cells and mediates downregulation of myeloid activity. In mice, it is found on monocytes, neutrophils, T-lymphocytes, and mast cells. Alternative splicing results in multiple transcript variants, and CD200R dysfunction is associated with primary hypertrophic osteoarthropathy.
