Product Specific Information

The HB15e monoclonal antibody reacts with human CD83, a 45 kDa transmembrane glycoprotein. CD83, a member of the Ig superfamily, is expressed on cultured dendritic cells, interdigitating, follicular, and circulating dendritic cells as well as some proliferating lymphocytes and human cell lines. While the function of CD83 is unclear, it serves as a useful marker for mature human blood dendritic cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of human peripheral blood-derived dendritic cells. Recommended ≤1 µg/test (in 100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

CD83 is a 40–45 kDa glycoprotein expressed by peripheral blood dendritic cells. Peripheral lymphocytes can express low levels of CD83 when cultured with agents such as Con A or PHA. In immunohistology, CD83 is strongly expressed by interfollicular interdigitating reticulum cells and weakly by germinal centre cells. It is also expressed by Langerhans cells in the skin.

The CD83 antigen is a 186-amino-acid single-chain glycoprotein, a member of the immunoglobulin superfamily, composed of an extracellular V-type Ig-like domain, a transmembrane region, and a short cytoplasmic tail (~40 amino acids). CD83 undergoes extensive post-translational glycosylation, resulting in a molecular weight approximately twice that of the core protein.

CD83⁺ cells display a unique surface phenotype distinct from T, B, NK, or myelomonocytic cells. They coexpress high levels of MHC class II molecules and markers such as CD2, CD5, CD40, CD78, CD13, CD33, CD36, as well as cytokine receptors and other surface molecules.
Diseases associated with CD83 dysfunction include plague and Rift Valley Fever.
