Product Specific Information
The IT2.2 monoclonal antibody reacts with human CD86 (B7-2), an ~80 kDa surface receptor belonging to the B7 family of costimulatory molecules. CD86 is expressed at low levels on B cells, macrophages, and dendritic cells, and is upregulated upon stimulation through the BCR complex, CD40, and cytokine receptors.
CD86 interacts with T-cell surface molecules CD28 and CTLA-4 (CD152), playing a critical role in T-B cell crosstalk, T-cell costimulation, and antibody production. Upregulation kinetics suggest its major role during the primary immune response phase.
Applications Reported: Flow cytometric analysis and functional blocking of CD86.
Applications Tested: Verified by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤0.25 µg/test (100 µL volume). Cell number may vary (10⁵–10⁸ cells/test). Antibody titration is advised for optimal performance.
Target Information
CD86 is a ligand for CTLA4 and CD28, acting as a costimulatory molecule in T-cell activation during antigen presentation. Insufficient co-stimulation can induce tolerance. CD86 is expressed by antigen-presenting cells and mediates both activation (via CD28) and inhibition (via CTLA4) of T-cell responses.
Alternative splicing produces multiple CD86 isoforms. Dysfunction in CD86 has been associated with gallbladder squamous cell carcinoma and myocarditis.
