Target Information

Programmed death-ligand 2 (PD-L2), also known as B7-DC, is a member of the B7 ligand family within the immunoglobulin superfamily. Along with PD-L1, it acts as a ligand for PD-1. PD-L2 is primarily expressed in dendritic cells but can be induced in various immune and non-immune cells depending on the microenvironment.

PD-L2 expression is upregulated by cytokines such as IL-4, TNF-alpha, and IFN-gamma. Although expressed at lower levels than PD-L1, PD-L2 shows a 2–6 times higher affinity for PD-1. Binding of PD-L2 or PD-L1 to PD-1 inhibits T cell activation, proliferation, and cytokine secretion, thus modulating immune responses.

Recent studies indicate that PD-L2 can also interact with RGMb (repulsive guidance molecule b), influencing respiratory tolerance via BMP and neogenin signaling pathways. The human PD-L2 protein includes a 201-amino-acid extracellular domain, a 21-amino-acid transmembrane domain, and a 32-amino-acid cytoplasmic domain.
