Target Information
BCL-2 is a key regulator of apoptosis that functions to either inhibit or promote cell death. Members of the BCL-2 family dimerize to modulate apoptosis. Bag1 forms a heterodimer with BCL-2 enhancing its anti-apoptotic effect. Bax and Bak are critical in cytochrome c release from mitochondria, initiating apoptosis. Constitutive expression of BCL2, such as in translocation to the Ig heavy chain locus, is associated with follicular lymphoma. Neoplastic germinal centers express high levels of BCL-2 alpha protein, while normal germinal centers are negative. Two transcript variants of BCL-2 differ at their C-terminal ends. Overexpression of BCL-2 is linked to cancers including B-cell lymphoma and prostate cancer.
