Product Specific Information
The MFL3 monoclonal antibody reacts with mouse Fas (CD95) Ligand, a 40 kDa type II transmembrane glycoprotein. FasL is a member of the TNF family and is expressed by activated mouse T cells. Interaction of FasL with its receptor CD95 induces Fas-mediated apoptosis.
Human FasL is known to be cleaved by matrix metalloproteinases (MMPs), producing a soluble 26 kDa form. Sensitivity of mouse FasL to MMPs has not been reported.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of transfected cells, ≤ 0.5 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.
Fluorochrome Information:
Super Bright 436 can be excited by violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.
For panels with multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information
CD178 (Fas ligand, FasL) is a type-II membrane protein with its N-terminus in the cytoplasm and C-terminal region extracellular. Its receptor, FasR, is a type-I membrane protein belonging to the TNF and NGF receptor family.
CD178 induces apoptosis upon FasR binding and exists in membrane-bound (45 kDa) or soluble (26 kDa) forms. Soluble FasL is released by metalloproteinase cleavage.
Binding of CD178 to Fas triggers receptor oligomerization and apoptosis. CD178 is mainly expressed in activated T lymphocytes and NK cells, and also in immune-privileged tissues (testis, eye). Expression has been reported in thymus, liver, ovary, lung, heart, and kidney.
CD178-mediated apoptosis is crucial in antiviral immune responses and regulation of immune reactions. Abnormal Fas/CD178 signaling is associated with diseases such as AIDS, hepatitis, cancer, atherosclerosis, and inflammatory conditions.
