Product Specific Information
Description:
The DTA-1 monoclonal antibody reacts with mouse GITR (Glucocorticoid-Induced TNFR family gene, TNFRSF18). In naive mice, GITR is expressed mainly by CD4⁺CD25⁺ T regulatory (Treg) cells and CD25⁺ CD4⁺ CD8⁻ thymocytes. Stimulation with DTA-1 abrogates Treg-mediated suppression. While it does not deplete CD357-expressing cells in vivo, it can induce tumorogenesis. Removal of GITR-expressing Treg cells or DTA-1 administration can lead to organ-specific autoimmune disease.

Target Information
Tumor necrosis factor receptor superfamily member 18 (TNFRSF18), also known as GITR or AITR, is encoded by the TNFRSF18 gene located on chromosome 1p36.33. It belongs to the TNF-receptor superfamily and is upregulated upon T-cell activation. This receptor is critical for maintaining immunological self-tolerance via CD25⁺CD4⁺ regulatory T cells. Mouse knockout studies indicate a role in regulating CD3-driven T-cell activation and apoptosis. Three alternatively spliced transcript variants encoding distinct isoforms have been reported.
