Product Specific Information

The monoclonal antibody 22C11 recognizes human, mouse, and rat APP (Amyloid Precursor Protein). APP is highly expressed in the central nervous system and exists in three major isoforms from alternative splicing. It is involved in synaptic formation, neuronal transport, iron export, and hormonal regulation. Secreted APP (sAPP) may provide neuroprotective effects against oxidative stress and excitotoxicity.

APP belongs to a family including APLP1 and APLP2, which share functional similarities. Proteolytic cleavage of APP produces beta amyloid, the main component of senile plaques—one of the hallmarks of Alzheimer’s disease. Dysregulation of APP processing is also implicated in Down’s syndrome, early onset familial Alzheimer’s disease, cerebral hemorrhage, and arthritis.

This antibody reacts with pre-A4 and recognizes all three isoforms of APP (immature, sAPP, and mature). It also cross-reacts with APLP2.

Applications Reported: Microscopy and immunocytochemistry.
Applications Tested: Tested by immunocytochemistry of methanol-fixed cells at ≤2.5 µg/mL. Optimal titration recommended.


Target Information

Amyloid Precursor Protein (APP) functions as a cell surface kinesin I membrane receptor, mediating axonal transport of beta-secretase and presenilin 1. It is a 100–140 kDa transmembrane glycoprotein with multiple isoforms. Proteolytic cleavage by beta- and gamma-secretases generates beta amyloid, the primary component of senile plaques—key features of Alzheimer’s disease. Abnormal APP regulation is linked to Down’s syndrome, familial Alzheimer’s, and cerebral hemorrhage.
