Product Specific Information

Description:
The monoclonal antibody V1NC3IDO recognizes human IDO (indoleamine-2,3-dioxygenase, INDO, IDO1), an intracellular enzyme that catalyzes the degradation of tryptophan to kynurenines. IDO is expressed in various tissues and cells, including macrophages, plasmacytoid dendritic cells, and several cell lines. It can be induced by IFN‑γ or other inflammatory stimuli. Expression of IDO in antigen-presenting cells and tumors is believed to mediate immune suppression via tryptophan depletion or production of metabolites that induce T‑cell apoptosis and regulatory T‑cell activation.

This antibody reacts with IDO1 and does not cross-react with IDO2.

Applications Reported:
Suitable for immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections and microscopy.

Applications Tested:
Validated for IHC‑P using low‑pH antigen retrieval at ≤ 0.5 µg/mL. Optimal dilution should be determined experimentally.

Target Information

IDO1 is an intracellular heme-containing enzyme that catalyzes the oxidative cleavage of the indole ring of regulatory molecules such as tryptophan, serotonin, and melatonin, leading to the production of kynurenines. It is widely expressed in human tissues, macrophages, and dendritic cells. During inflammation, IFNs induce IDO1 expression, contributing to antimicrobial defense by depleting tryptophan and inhibiting pathogen growth. IDO1 also plays crucial roles in immunoregulation during infection, pregnancy, autoimmunity, transplantation, and cancer.
