The A6.1 monoclonal antibody recognizes human, mouse, and rat thrombospondin-1, a member of the thrombospondin family of extracellular calcium-binding proteins.
Thrombospondin-1 is a secreted glycoprotein originally discovered in activated platelets but also expressed by endothelial cells, adipocytes, fibroblasts, macrophages, monocytes, and smooth muscle cells.
It binds extracellular matrix components such as fibronectin, integrins, and heparin, with signaling mediated through the TGF beta pathway.
Thrombospondin-1 is involved in angiogenesis, inflammation, osteogenesis, cell proliferation, and synaptogenesis during neural development.

Applications Reported: Immunoprecipitation, Western blotting, immunohistochemistry (formalin-fixed paraffin-embedded tissue), microscopy, ELISA, and immunocytochemistry.
Applications Tested:

• IHC (paraffin): ≤ 1 µg/mL using high pH antigen retrieval
• ICC: ≤ 1 µg/mL with formaldehyde or methanol fixation
  Careful titration is recommended for optimal assay performance.

Target Information

Thrombospondin is a protein from platelet α-granules composed of three identical subunits bound by interchain disulfides. It is secreted at sites of platelet activation and aggregation and participates in chemotaxis, adhesion, proliferation, and differentiation of leukocytes, fibroblasts, smooth muscle, and endothelial cells.
