Target Information

The WISP1 gene encodes a member of the WNT1 inducible signaling pathway (WISP) subfamily, part of the connective tissue growth factor (CTGF) family. WNT1 proteins are cysteine-rich, glycosylated signaling molecules involved in diverse developmental processes. CTGF family members contain four conserved cysteine-rich domains: insulin-like growth factor-binding domain, von Willebrand factor type C module, thrombospondin domain, and a C-terminal cystine knot-like domain.

WISP1 may act downstream in the WNT1 signaling pathway relevant to malignant transformation. It is highly expressed in fibroblast cells and overexpressed in colon tumors. The encoded protein binds to decorin and biglycan—small leucine-rich proteoglycans in the extracellular matrix—potentially inhibiting their suppression of tumor cell proliferation. WISP1 also attenuates p53-mediated apoptosis via Akt kinase activation. It shares 83% amino acid identity with the mouse protein. Two transcript variants are generated by alternative splicing.
