Description:
This BCL-DWN monoclonal antibody reacts with mouse Bcl-6 (B-cell lymphoma 6), a member of the POZ family of transcriptional repressors. Bcl-6 is abundantly expressed in germinal center B cells and in differentiating/activated B cells. The 79-kDa protein is essential for germinal center B cell proliferation and survival, and supports pre-B cell survival during development.
Bcl-6 is constitutively expressed in diffuse large B cell lymphomas (DLBCLs), Burkitt’s lymphoma, and non-Hodgkin’s lymphoma, and has been reported in CD4+ T follicular helper and memory T cells. It is antagonized by Blimp-1.

Reported for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Tested by intracellular staining and flow cytometric analysis of splenocytes from sheep red blood cell-immunized mice using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Refer to BestProtocols®: Protocol B for intracellular nuclear proteins.
Use ≤ 0.03 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirically determined). Antibody titration recommended for optimal results.

Target Information
Bcl-6 proto-oncogene product is a Kruppel-type zinc-finger protein mainly expressed in germinal center B cells and related lymphomas. It is involved in chromosome rearrangements at 3q27 in non-Hodgkin’s lymphomas, with rearrangements detected in 33–45% of diffuse large B cell lymphomas. Bcl-6 is also detected in follicular lymphomas, diffuse large B cell lymphomas, Burkitt’s lymphomas, and nodular lymphocyte-predominant Hodgkin’s disease.
