Target Information
The KRAB (Kruppel-associated box) domain, approximately 45 amino acids in length, is a transcriptional repression domain found in numerous transcription factors. Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes have been identified in the human genome.
These proteins repress transcription via interactions with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing RING finger, B boxes, and PHD finger domains. KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency and interacts directly with HP1 (heterochromatin protein 1) and KRAZ1 (Kruppel-associated box-containing zinc finger protein 1). KAP-1 targets KRAZ1 to centromeric heterochromatin regions containing HP1alpha, thereby regulating transcriptional repression. Mutant KAP-1 unable to bind HP1 leads to random KRAZ1 distribution and strong transcriptional activation.
