이 항체는 Western blot, immunofluorescence, immunohistochemistry, immunoprecipitation에 성공적으로 사용되었습니다.
- Western blot에서 비환원 조건에서는 25 kDa (pentameric PLB), 환원 조건에서는 5 kDa (monomeric PLB)를 검출합니다.
- Rat papillary muscle에서 면역형광 염색 시 sarcoplasmic reticulum(SR)의 네트워크에 확산된 염색 패턴을 보입니다.
- Ventricular SR vesicle의 칼슘 흡수를 증가시켜 PLB의 인산화 상태를 모방할 수 있습니다.
- 면역형광 염색용으로 권장됩니다.
Immunogen은 canine PLB의 잔기 D(2)–R(25)에 해당하는 합성 펩타이드이며, 이 항체는 canine PLB의 아미노산 9–17 사이의 epitope을 인식합니다.
Target Information
Phospholamban (PLB)은 52개의 아미노산으로 구성된 인산화 단백질로, 심근 세포의 sarcoplasmic reticulum(SR) 내 칼슘 펌프를 조절합니다.
PLB는 5개의 동일한 서브유닛으로 이루어진 올리고머로, 각 서브유닛은 세포질 도메인(1–25)과 막관통 도메인(25–52)을 가집니다.
세포질 도메인은 인산화 부위를 포함하며 단백질분해효소에 민감하고, 막관통 도메인은 소수성이며 pentamer 구조를 안정화시킵니다.
