MA3-916은 ryanodine receptor 2 (RyR2)를 검출하며, amphibian, canine, chicken, fish, guinea pig 및 rat 조직에서 RyR1과 교차 반응성을 보입니다.
이 항체는 Western blot, immunohistochemistry, immunofluorescence, immunocytochemistry, immunoprecipitation에 성공적으로 사용되었습니다.
Western blot에서 565 kDa 단백질 밴드를 검출하며, 이는 ryanodine receptor를 나타냅니다. 비포유류 척추동물에서는 565 kDa 부근에서 α 및 β isoform을 나타내는 doublet이 관찰됩니다.
Rat 심근조직에서의 면역염색 결과는 근섬유 내 강한 염색으로 나타나며, 이는 sarcoplasmic reticulum에 국한된 발현 양상과 일치합니다.
Target Information
Ryanodine receptor (RyR)는 근세포의 sarcoplasmic reticulum(SR)에서 칼슘 방출을 담당하는 채널로, 비근육세포에서도 칼슘 조절에 관여합니다.
RyR은 homotetramer 형태로 존재하며, 짧은 세포질 C-말단과 4~10개의 막관통 도메인을 가집니다.
단백질의 나머지 부분은 T-tubule과 SR 사이의 세포질 내 foot 영역에 위치합니다.
포유류에서는 세 가지 유전자형이 존재합니다:
- RyR-1: 주로 골격근 및 뇌 일부 영역에서 발현
- RyR-2: 주로 심근에서 발현되며, 위, 내피세포, 뇌 일부에서도 존재
- RyR-3: 평활근 및 뇌(선조체, 시상, 해마 등)에서 발현
