Target Information

Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
This protein is a key component of the extracellular matrix in cartilage, providing resistance to compression.
Human Aggrecan consists of 2316 amino acids (~250 kDa) and exists in multiple isoforms due to alternative splicing.

Aggrecan has a bottlebrush structure with chondroitin sulfate and keratan sulfate chains attached to its core.
Its synthesis and degradation are closely related to cartilage deterioration during joint injury, disease, and aging.

It contains three interglobular domains (IGDs)—G1, G2, and G3—that are involved in:

• Aggregation
• Hyaluronan binding
• Cell adhesion
• Chondrocyte apoptosis

Aggrecan fragments are released into synovial fluid during all stages of osteoarthritis due to cleavage by aggrecanase and MMP activity.
Antibodies BC-3 and BC-4 can detect neo epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
