Target Information

Glutamate is the main excitatory neurotransmitter in the brain. Initially, it was known to act only on ligand-gated receptor channels such as NMDA, AMPA, and kainate receptors, which mediate fast excitatory synaptic transmission.
Recent studies show that glutamate also regulates enzymes producing second messengers via specific G-protein-coupled receptors known as metabotropic glutamate receptors (mGluRs).

  • Group I receptors (mGluR1a, mGluR5): Stimulate phospholipase C, increasing phosphoinositide turnover and calcium release.
  • Group II and III receptors (mGluR2, mGluR3): Inhibit adenylyl cyclase.
