Target Information
Glutamate is the main excitatory neurotransmitter in the brain. It acts through ligand-gated receptor channels such as NMDA, AMPA, and kainite receptors involved in fast excitatory synaptic transmission.
Recently, glutamate has been shown to regulate enzymes producing second messengers via specific receptors coupled to G-proteins, known as metabotropic glutamate receptors (mGluRs).
Group-I receptors (mGluR1a, mGluR5) stimulate phospholipase C, increasing phosphoinositide turnover and calcium release from internal stores.
Group-II and -III receptors (including mGluR2 and mGluR3) are coupled to the inhibition of adenylyl cyclase.
