Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C followed by cooling at room temperature for 20 minutes.
Target Information
NPM1 (Nucleophosmin 1, B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, cytoplasmic/nuclear trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and regulation of p53.
It shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones. NPM1 binds nucleic acids, prevents protein aggregation, and protects enzymes during thermal denaturation.
It is associated with centrosome duplication as a substrate of CDK2/cyclin E. NPM1 participates in fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, contributing to oncogenic processes.
Dysfunction of NPM1 is linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis.
