Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, then cool at room temperature for 20 min.

Target Information

NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in:

  • Ribosome assembly and transport
  • Cytoplasmic/nuclear trafficking
  • Regulation of DNA polymerase α activity
  • Centrosome duplication
  • Regulation of p53

NPM1 shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones. It binds nucleic acids, prevents protein aggregation, protects enzymes during thermal denaturation, and assists protein renaturation. NPM1 is associated with centrosome duplication and acts as a substrate of CDK2/cyclin E. It interacts with tumor-associated translocations forming fusion proteins (NPM-ALK, NPM-RAR, NPM-MLF1) and may facilitate oncogenic conversion. Additionally, NPM1 sequesters tumor suppressor RF in the nucleolus, protecting it until required. Dysfunction of NPM1 is linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis.
