Product Specific Information
- Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
- For formalin-fixed tissues, heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, then cool at room temperature for 20 minutes.
Target Information
NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in:
- Ribosome assembly and transport
- Cytoplasmic/nuclear trafficking
- Regulation of DNA polymerase α activity
- Centrosome duplication
- Regulation of p53
NPM1 shuttles continuously between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. It binds nucleic acids, prevents protein aggregation, protects enzymes during thermal denaturation, and assists in renaturation of denatured proteins.
NPM1 is associated with centrosome duplication and is a substrate of CDK2/cyclin E. It is implicated in several tumor-associated fusion proteins (NPM-ALK, NPM-RAR, NPM-MLF1), acting as an oligomerization interface.
Additionally, NPM1 sequesters tumor suppressor proteins in the nucleolus, protecting them from degradation. Dysfunction of NPM1 is linked to diseases such as acute myeloid leukemia and lymphomatoid papulosis.
