Target Information

NPM1 (Nucleophosmin 1, also known as B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, nucleocytoplasmic trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and p53 regulation. It shuttles between the nucleus, cytoplasm, and nucleolus, acting as a chaperone for core histones. NPM1 binds nucleic acids, prevents protein aggregation, protects enzymes from thermal denaturation, and assists in protein renaturation. It is associated with centrosomes and serves as a substrate of CDK2/cyclin E during centrosome duplication. NPM1 is implicated in several tumor-associated fusion proteins (e.g., NPM-ALK, NPM-RAR, NPM-MLF1) and may contribute to oncogenic transformation. Dysregulation of NPM1 is linked to acute myeloid leukemia and lymphomatoid papulosis.
