Product Specific Information
- Antibody tested in direct ELISA (use at 2–4 µg/mL for coating).
- For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- For formalin-fixed tissues: heat sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, then cool at room temperature for 20 min.
Target Information
NPM1 (Nucleophosmin 1, B23, nutramin, NO38) is a ubiquitously expressed phosphoprotein involved in ribosome assembly and transport, cytoplasmic/nuclear trafficking, regulation of DNA polymerase alpha activity, centrosome duplication, and p53 regulation.
It shuttles between the nucleus, cytoplasm, and nucleolus, chaperoning core histones. NPM1 binds nucleic acids, prevents protein aggregation, protects enzymes during thermal denaturation, and aids in renaturation of chemically denatured proteins.
NPM1 interacts with tumor-associated chromosome translocations forming fusion proteins such as NPM-ALK, NPM-RAR, and NPM-MLF1, acting as an interface for oligomerization and oncogenic conversion.
Dysfunction of NPM1 is associated with diseases including acute myeloid leukemia and lymphomatoid papulosis.
