Product Specific Information

This 2F1 monoclonal antibody reacts with mouse Killer cell Lectin-like Receptor G1 (KLRG1), also known as Mast cell Function-associated Antigen (MAFA).
KLRG1 is a homodimer of glycosylated 30–38 kDa subunits containing a cytoplasmic motif similar to ITIM. Expression is found on approximately one-third of mouse NK cells and a subset of T cells.
MHC class I molecules regulate KLRG1 through Ly49 and SHP-1 signaling.

Applications Reported: Flow cytometric analysis.
Applications Tested: Mouse spleen cells, ≤0.125 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with 488 nm blue laser.
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Light-sensitive—protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)
  Performance after fixation may vary; determine empirically.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

KLRG1 (Killer cell lectin-like receptor G1, MAFA, 2F1-Ag) is a homodimeric lectin-like type 2 transmembrane receptor containing ITIM motifs that interact with SH2 domains of phosphatases such as SHP-1.
It acts as an inhibitory receptor expressed on NK cells and certain T cells, involved in tumor and virus-infected cell lysis and immune regulation.
