Target Information
DNA-directed RNA polymerase II subunit RPB1 (POLR2A) is a DNA-dependent RNA polymerase that catalyzes transcription of DNA into RNA using ribonucleoside triphosphates as substrates. POLR2A is the largest subunit and a catalytic component of RNA polymerase II, synthesizing mRNA precursors and non-coding RNAs. It forms the polymerase active center with the second largest subunit and is central to the basal transcription machinery.
POLR2A includes mobile structural elements such as the clamp and jaws that interact with the DNA template. The bridging helix from RPB1 promotes translocation of Pol II by conformational changes during nucleotide addition. Transcription elongation is regulated by the phosphorylation of the CTD of RPB1, serving as a scaffold for factors controlling initiation, elongation, termination, and mRNA processing. Gene expression regulation depends on the balance of CTD-lysine methylation and acetylation.
