The 8D4-8 antibody reacts with human interleukin-4 (IL-4), a 15–19 kDa cytokine secreted by Th2 cells.
- Applications Reported: Used as a capture antibody for human IL-4 ELISA and for intracellular staining in flow cytometric analysis.
- Applications Tested: Tested in intracellular staining and flow cytometry of stimulated human peripheral blood cells. Recommended ≤0.125 µg/test (100 µL final volume). Optimal titration required.

Target Information
Interleukin-4 (IL-4) is a pleiotropic, immune-modulatory cytokine produced by activated T cells and acts as a ligand for the IL-4 receptor. IL-4 and IL-13 share overlapping functions through receptor binding. STAT6 mediates IL-4 signaling. The IL-4 gene is located on chromosome 5q within a cytokine gene cluster (IL-3, IL-5, IL-13, CSF2). IL-4, IL-13, and IL-5 are co-regulated by long-range elements over a 120 kb region. IL-4 delta 2 is a known splice variant.
