The 5D3-F7 antibody reacts with human, cynomolgus, and rhesus monkey monocyte chemoattractant protein-1 (MCP-1), also known as monocyte chemotactic and activating factor (MCAF) and CCL2. MCP-1 is a CC chemokine that forms a dimer and acts as a chemoattractant for monocytes, basophils, T cells, and NK cells through receptors such as CCR2, CCR5, and CCR10.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Tested by intracellular staining and flow cytometry using stimulated human peripheral blood cells with the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00) and BestProtocols® Protocol A.
• Recommended usage ≤ 0.06 µg/test (in 100 µL final volume).
• Cell number: 10⁵–10⁸ cells/test (empirically determined).
• Antibody titration recommended for optimal performance.

Target Information

This gene encodes a cytokine located on chromosome 17q, belonging to the CC chemokine family. It has chemotactic activity for monocytes and basophils, but not for neutrophils or eosinophils. It is implicated in inflammatory diseases such as psoriasis, rheumatoid arthritis, and atherosclerosis, and binds to chemokine receptors CCR2 and CCR4.
