Product Specific Information

This antibody reacts with human p53 tumor suppressor intracellular protein phosphorylated at CKII site (Ser392).
For IHC staining, high temperature antigen retrieval (microwave or pressure cooker) is recommended, using 10 mM citrate buffer (pH 6.0) or 1 mM EDTA-NaOH buffer (pH 8.0).

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation.
p53 up-regulates transcription of genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, p53 contains:

  • N-terminal transactivation domain
  • Central DNA-binding domain
  • Oligomerization domain
  • C-terminal regulatory domain

Phosphorylation at Ser15 and Ser392 plays key roles in activation and tumor association. Mutations in the TP53 gene are common in cancers and cause loss of tumor suppressor activity.
Alterations of TP53 also occur as germline mutations in cancer-prone families (e.g., Li-Fraumeni syndrome).
p53 is expressed at low levels in normal cells but highly in transformed cells, contributing to malignancy, aging, and oxidative stress responses.
