Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. It promotes tubulin polymerization and stabilizes microtubules. The C-terminus binds axonal microtubules while the N-terminus interacts with neural plasma membrane components, functioning as a linker between both structures. Tau localization determines axonal polarity within neurons.

Short isoforms provide cytoskeletal plasticity, whereas longer isoforms contribute to stabilization. In its hyper-phosphorylated form, Tau constitutes the major component of paired helical filaments (PHF), which form neurofibrillary lesions characteristic of Alzheimer’s disease (AD). Hyper-phosphorylation reduces microtubule binding, leading to destabilization and neuronal degeneration. Multiple kinases, including GSK-3β, PKA, cdk5, and casein kinase II, phosphorylate Tau. Hyper-phosphorylated Tau is also implicated in other neurodegenerative disorders such as Pick’s disease, frontotemporal dementia, corticobasal degeneration, and progressive supranuclear palsy.
