Target Information

The KRAB (Kruppel-associated box) domain is approximately 45 amino acids long and functions as a transcriptional repression domain found in many transcription factors. Over 220 KRAB-zinc finger protein (KRAB-ZFP) genes have been identified in the human genome.
These proteins repress transcription through interactions with KAP-1 (KRAB-associated protein 1), an 835 amino acid polypeptide containing a RING finger, B boxes, and a PHD finger. KAP-1 forms complexes with KRAB-domain transcription factors to enhance repression efficiency.
KAP-1 interacts directly with HP1 (heterochromatin protein 1) and KRAZ1 (Kruppel-associated box-containing zinc finger protein 1), targeting KRAZ1 to centromeric heterochromatin foci containing HP1α, thereby regulating transcriptional repression.
Mutant KAP-1 proteins that bind KRAB but not HP1 result in random KRAZ1 distribution and strong transcriptional activation.
