Product Specific Information

The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. It is expressed mainly on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are known ligands for PD-1, showing overlapping functions with constitutive expression on some normal tissues and upregulation on activated antigen-presenting cells.

J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo, J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes, similar to PD-1-deficient cells.

Target Information

T-cell activation is self-limited by inhibitory receptors such as PD-1 (PDCD-1), which binds ligands PD-L1 and PD-L2 on antigen-presenting cells. Upon ligand binding, PD-1 transmits inhibitory signals that suppress immune activation in the absence of danger signals. PD-1 deficiency leads to hyperactivated immune responses and autoimmune diseases. Although its predicted molecular weight is 55 kDa, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
