Product Specific Information

The MIH4 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains an immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. It is predominantly expressed on activated T and B lymphocytes.

PD-L1 (B7-H1) and PD-L2 (B7-DC) are PD-1 ligands, showing overlapping functions and expression on normal tissues and activated antigen-presenting cells.
The MIH4 antibody recognizes a different epitope than clone J105.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on PHA-stimulated human peripheral blood cells.
Use at 5 µL (1 µg) per test in a final volume of 100 µL (10⁵–10⁸ cells/test).

Target Information

PD-1 (PDCD-1) is an inhibitory receptor expressed on activated T cells, regulating immune responses by binding to ligands PDL-1 and PDL-2 on antigen-presenting cells. Binding inhibits immune activation in the absence of danger signals (e.g., LPS). PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune disease. Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
