Description:
The 7E.17G9 monoclonal antibody reacts with mouse ICOS (Inducible COStimulatory molecule), a T cell-specific molecule and a third member of the CD28/CTLA-4 family. ICOS is a 47–57 kDa homodimer expressed on activated T cells, playing a key role in costimulation for T cell activation and proliferation, and is required for humoral immune responses. It binds to its ligand B7h/B7RP-1 on activated APCs, including B cells, and may have a protective role in inflammatory autoimmune diseases. ICOS may also be involved in Th2 cell development.

This 7E.17G9 antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of stimulated mouse splenocytes. Use ≤0.125 µg per test (100 µL final volume). Cell numbers may vary (10⁵–10⁸ cells/test). Optimize titration for best results.

Fluorochrome Information:
PerCP-eFluor™ 710 emits at 710 nm and is excited by the 488 nm blue laser. It can substitute for PerCP-Cyanine5.5. Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal loss of brightness or FRET efficiency. Clone-specific performance should be validated empirically.

Excitation / Emission / Laser:
488 nm excitation; 710 nm emission; Blue laser.
