Product Specific Information

The MIH4 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the immunoglobulin superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. It is predominantly expressed on activated T and B lymphocytes. PD-L1 (B7-H1) and PD-L2 (B7-DC) are identified ligands of PD-1, both showing overlapping functions and expression patterns.

This antibody recognizes a different epitope than clone J105.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on stimulated human peripheral blood cells at 5 µL (1 µg)/test.
A test is defined as the amount of antibody staining a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

Fluorochrome Information:

  • PerCP-eFluor® 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5.
  • Excitation: 488 nm; Emission: 710 nm; Laser: Blue Laser.
  • Recommended filters: 685 LP dichroic mirror and 710/40 band pass filter (or 695/40 alternative).
  • Stable in 2% formaldehyde overnight at 4°C.
  • Filtration: 0.2 µm post-manufacturing filtered.

Target Information

PD-1 (PDCD-1) is a receptor expressed on activated T cells that mediates inhibitory signals via ligands PDL-1 and PDL-2. Binding of these ligands downregulates immune activation in the absence of danger signals such as LPS. PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune diseases. Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
